Background
Vascular endothelial growth factor A (VEGF-A/VEGF165) is the prototypical pro-angiogenic member of the VEGF/PDGF growth factor family and a central regulator of both physiological and pathological angiogenesis. The 165-amino-acid isoform is the predominant secreted species; it forms a disulfide-linked homodimer that signals through VEGFR1 (Flt-1), VEGFR2 (KDR/Flk-1) and co-receptor neuropilin-1 on endothelial cells, driving proliferation, migration, survival and vascular permeability. Hypoxia and oncogenic signaling potently induce VEGF-A, which tumors exploit to build their blood supply — a process targeted clinically by bevacizumab and by the anti-VEGF biologics ranibizumab, aflibercept and faricimab used in wet age-related macular degeneration and other retinal vascular diseases. The human sequence is identical in cynomolgus macaque, so this protein serves cross-species (NHP) pharmacology studies directly. Bioactive recombinant VEGF165 is a workhorse reagent for endothelial culture, angiogenesis models and potency assays of anti-VEGF therapeutics.
Formulation & Handling
- Formulation
- Supplied in either a lyophilized powder or a ready-to-use liquid format. Buffer: PBS, pH 7.4, with 5% trehalose as cryoprotectant.
- Reconstitution
- Lyophilized: briefly centrifuge the vial before opening, reconstitute in sterile distilled water to 0.1–1.0 mg/mL, and let stand at room temperature for 10 minutes before aliquoting. Liquid: ready to use — thaw on ice and equilibrate.
- Storage
- Both lyophilized and liquid proteins are stable for 12 months at -40 °C to -80 °C. After reconstitution, aliquot and store at -40 °C to -80 °C. Avoid repeated freeze-thaw cycles.
- Shipping
- Shipped on dry ice or cold packs.
