Bioactivity
Measured by functional ELISA. Immobilized recombinant human c-MET ECD binds recombinant human HGF with an EC50 in the ng/mL range; binding is blocked by reference anti-c-MET antibodies.
Background
c-MET (hepatocyte growth factor receptor, HGFR) is a receptor tyrosine kinase whose misregulation — exon 14 skipping, amplification or overexpression — is a defined oncogenic driver in NSCLC, gastric cancer and other malignancies. Normally, HGF binding to the extracellular SEMA and PSI domains triggers scattering, invasion and branched morphogenesis during tissue repair; in cancer the same program becomes invasive growth and therapy resistance. The MET arena now spans highly selective TKIs (capmatinib, tepotinib, savolitinib), MET-directed antibodies and antibody–drug conjugates, all of which require reliable extracellular-domain reagents for ligand-blocking, receptor occupancy and pharmacodynamic assays. This ECD construct covers the full HGF-binding ectodomain for those applications.
Formulation & Handling
- Formulation
- Supplied in either a lyophilized powder or a ready-to-use liquid format. Buffer: PBS, pH 7.4, with 5% trehalose as cryoprotectant.
- Reconstitution
- Lyophilized: briefly centrifuge the vial before opening, reconstitute in sterile distilled water to 0.1–1.0 mg/mL, and let stand at room temperature for 10 minutes before aliquoting. Liquid: ready to use — thaw on ice and equilibrate.
- Storage
- Both lyophilized and liquid proteins are stable for 12 months at -40 °C to -80 °C. After reconstitution, aliquot and store at -40 °C to -80 °C. Avoid repeated freeze-thaw cycles.
- Shipping
- Shipped on dry ice or cold packs.
