Bioactivity
Measured by functional ELISA. Immobilized recombinant human factor IX binds reference anti-factor IX antibodies with EC50 values in the ng/mL range.
Background
Coagulation factor IX is a vitamin K-dependent serine protease zymogen of the intrinsic pathway; its genetic deficiency causes hemophilia B, one of the classic inherited bleeding disorders. The mature protein organizes a Gla domain (γ-carboxylated for membrane binding), two EGF-like domains, an activation peptide and a catalytic protease domain — a modular architecture that has made FIX a flagship of modern hemophilia care, from engineered replacement products and gene therapy (Octaplas/fidanacogene elaparvovec lineage) to bispecific antibodies mimicking its cofactor function. Mammalian-cell-expressed recombinant factor IX preserving γ-carboxylation supports inhibitor-antibody assays, pharmacokinetic research and cascade modeling.
Formulation & Handling
- Formulation
- Supplied in either a lyophilized powder or a ready-to-use liquid format. Buffer: PBS, pH 7.4, with 5% trehalose as cryoprotectant.
- Reconstitution
- Lyophilized: briefly centrifuge the vial before opening, reconstitute in sterile distilled water to 0.1–1.0 mg/mL, and let stand at room temperature for 10 minutes before aliquoting. Liquid: ready to use — thaw on ice and equilibrate.
- Storage
- Both lyophilized and liquid proteins are stable for 12 months at -40 °C to -80 °C. After reconstitution, aliquot and store at -40 °C to -80 °C. Avoid repeated freeze-thaw cycles.
- Shipping
- Shipped on dry ice or cold packs.
