Recombinant Human SOST/Sclerostin Protein (C-His)

Measured by functional ELISA. Immobilized recombinant human sclerostin binds LRP6 and reference anti-sclerostin antibody (romosozumab class) with EC50 values in the ng/mL range.

Sclerostin (SOST) is a secreted glycoprotein made almost exclusively by osteocytes, where it acts as the master brake on bone formation: by binding the Wnt co-receptors LRP5/LRP6 it shuts down canonical Wnt signaling in osteoblasts, and loss-of-function mutations in humans produce the high-bone-mass phenotypes sclerosteosis and van Buchem disease. That biology made sclerostin inhibition the first true bone-anabolic approach beyond PTH — romosozumab blocks sclerostin to build bone rapidly in postmenopausal osteoporosis. Recombinant human sclerostin is the central reagent for osteoanabolic drug screening, Wnt-axis binding assays and bone-metabolism assay standards.
