Bioactivity
Measured by functional ELISA. Immobilized recombinant human TROP-2 binds reference anti-TROP-2 antibody (sacituzumab class) with an EC50 in the range of 10–50 ng/mL.
Background
TROP-2 (trophoblast cell-surface antigen 2, TACSTD2) is a cell-surface glycoprotein of the trophoblast cell-surface antigen family that regulates calcium signaling and cell-cycle progression, and is overexpressed across epithelial malignancies — most prominently triple-negative and HR+/HER2- breast, urothelial, gastric and lung cancers, where high expression correlates with poor prognosis. Its dense, broadly retained tumor expression with limited normal-tissue presence made TROP-2 the leading ADC antigen of the current generation: sacituzumab govitecan established the target in breast and urothelial cancer, and datopotamab deruxtecan is advancing through late-stage trials. Recombinant TROP-2 extracellular domain is the core reagent for ADC screening, biosimilar potency testing and epitope characterization in this fast-moving arena.
Formulation & Handling
- Formulation
- Supplied in either a lyophilized powder or a ready-to-use liquid format. Buffer: PBS, pH 7.4, with 5% trehalose as cryoprotectant.
- Reconstitution
- Lyophilized: briefly centrifuge the vial before opening, reconstitute in sterile distilled water to 0.1–1.0 mg/mL, and let stand at room temperature for 10 minutes before aliquoting. Liquid: ready to use — thaw on ice and equilibrate.
- Storage
- Both lyophilized and liquid proteins are stable for 12 months at -40 °C to -80 °C. After reconstitution, aliquot and store at -40 °C to -80 °C. Avoid repeated freeze-thaw cycles.
- Shipping
- Shipped on dry ice or cold packs.
